Description

Glen UnySupport™ is a universal support based on a conformationally preorganized molecule to accelerate the dephosphorylation reaction. The N-phenyl version, developed at Isis Pharmaceuticals as UnyLinker™, is available from several companies for large scale oligonucleotide synthesis. Glen UnySupport™ is the N-methyl version, which is preferred for high throughput oligonucleotide synthesis since methylamine rather than aniline is formed on deprotection. Higher angstrom universal supports are ideal for long syntheses (>150 mer) and can be utilized with modifier phosphoramidites for which 2000Å supports are not readily available.

Usage

  • Coupling: This support should be used in a manner identical to normal protected nucleoside support since it contains the DMT group.
  • Deprotection: Solution phase: Ammonium Hydroxide:MethylAmine (AMA) 1:1 for 1 hour at 65°C or Ammonium Hydroxide for 8 hours at 55°C. For sensitive minor bases or dyes, Glen UnySupport may be eliminated with 50 mM Potassium Carbonate in Methanol in 17 hours at room temperature or with Tert-Butylamine/w
