Gene Expression Analysis
The products in this section are used in gene expression analysis. Specific applications include generation of ds DNA from small RNAs for high through-put (next-gen) sequencing templates, labelled small RNA for microarray analyses, identification of 5'' and 3'' eukaryotic mRNA, identification of exact transcription start sites and creation of cDNA libaries that represent full-length selected transcripts from total RNA or enriched mRNA. The TargetAmp Kits produce microgram amounts of aRNA form mRNA in total RNA and can incorporate both aminoallyl and biotin labels if desired.
TargetAmp aRNA Amplification
DNA microarrays provide a powerful method to measure the abundance of thousands of different RNA molecules in a single experiment. The amount of RNA required for gene expression analysis, however, can be a limiting factor when the available biological sample is small, as in the case of needle biopsies or laser capture micro-dissected cells. RNA amplification technology has made transcription profiling of such small samples feasible.
TargetAmp aRNA kits utilises a significantly improved 'Eberwine-type' aRNA amplification protocol, that generates microgram quantities of amplified antisense RNA from picogram amounts of total RNA. This represents at least 5,000,000-fold amplification of the polyadenylated component of the original total RNA, making microarray analysis of single cells possible.
The aRNA produced can be labeled with biotin or fluorescent dyes, and performs well on multiple microarray.
