Description

CviJ I is the only available restriction enzyme recognising two/three basepair sequence, thus allowing for new research applications.

This version of CviJ I cleaves only PuGCPy sites. It does not exhibit ‘star’ activity, thus it is better suited for mapping of short DNAs like PCR products or small plasmids. Other CviJ I applications, like shotgun cloning, thermal cycle labelling or epitope mapping, can be performed either with this version or with CviJ I*, also available from Cambio (Call for details).

One unit is defined as the amount of enzyme required to completely digest 1µg of pBR322 DNA in 1 hour in a total reaction volume of 25µl.

 

 

Assay Conditions 20mM Glycylglycine-KOH (pH 8.5)
0.1mM DTT
50mM K-acetate
10% DMSO
10mM Mg-acetate
1µg pBR322 DNA
Reaction volume 25µl
DMSO is supplied separately.
Storage Buffer 20mM Tris-acetate (pH 8.0)
50mM potassium acetate
0.5mM EDTA
1mM DTT
5mM MgCl2
50% (v/v) glycerol

 

Store enzyme at -80°C, buffer at -20°C and DMSO at Room Temp
