2’-OMe-N6-Me-A, commonly referred to as m6Am, is a
crucial component of the 5’-cap of mRNA. 2’-OMe-A is often the first
transcribed nucleotide, and this modification is a result of methylation
at the N6-position of the nucleobase. The 5’-cap structures initiate
translation and protect RNA from degradation. Internal m6Am,
have also been implicated in regulating gene expression, RNA splicing,
and cancer proliferation. We offer the 2’-OMe-N6-Me-A RNA monomer with a
phenoxyacetyl protecting group to minimize potential branching.
2’-OMe-N6-Me-A-CE Phosphoramidite is compatible with popular synthesis
and deprotection methods, from UltraMild to AMA.

Usage
- Coupling:
6 minute coupling time. To avoid any exchange of the iPr-Pac group on
the G with acetyl, use the UltraMild Cap Mix A (40-4210-xx/ 40-4212-xx).
- Deprotection:
UltraMild deprotection: 0.05M Potassium Carbonate in Methanol, 4 hours
at Room Temperature OR 2 hours at Room Temperature in 30% Ammonium
Hydroxide.
