Shark Lab: Great White Migration

Scientists have spotted
similar-looking shark fins in four very different locations–are white
sharks migrating across oceans? Students use DNA fingerprinting to
explore shark ecology in this engaging gel electrophoresis activity.
Are white sharks migrating across oceans? In this lab, students join a
team of scientists who have noticed a similar-looking shark fin off the
coasts of Australia, California, Hawai’i, and South Africa. Using DNA
fingerprinting, students determine whether an individual shark is
migrating across oceans to reach these different coastal waters. This
gel electrophoresis lab provides an introduction to DNA-based
identification in an ecology context. Perfect for introductory high
school or advanced middle school classes, this activity explores the
importance of white sharks in marine ecosystems and how scientific
research can inform conservation efforts.

What is included
Reagents for 8 lab groups:
- 4 simulated shark DNA samples for fingerprinting analysis
- Samples come premixed with gel loading dye
Storage notes:

- If kept in the refrigerator, reagents can be stored for 6 months after receipt.
Required equipment:
Gel electrophoresis reagents and plastic tubes are supplied by the user
- Agarose
- Gel electrophoresis buffer (e.g., TBE)
- Fluorescent DNA stain (e.g., SeeGreen™ or GelGreen®)
- Plastic tubes for distributing samples to student groups
The Learning Lab Companion Kit (KT-1510-01) provides sufficient reagents to prepare and run eight gels when using the blueGel™ or Bandit™ electrophoresis systems, as well as plastic tubes to distribute samples to student groups.
