Our supplier, LGC Biosearch Technologies, operates a dedicated production line with an optimized workflow specifically engineered to deliver oligos with high purity, excellent reproducibility, and maximal cross-contamination mitigation. Their NxGrade oligo production line is routinely monitored using NGS-based testing to help ensure effective cross-contamination control.

Low oligo cross-contamination in NGS indexing primers and adapters is essential for reliable sequencing results because even trace amounts of the wrong index can cause barcode misassignment, where reads are assigned to the wrong sample. Cross-contaminated or low-purity oligos, containing truncated sequences, can contribute to adapter crosstalk or index hopping. These issues can degrade data integrity, compromise downstream analysis, cause sample misidentification, waste sequencing capacity, delay projects, and increase overall costs.

Traditional HPLC purification methods reliably generate high-purity, full-length oligos, but they can also introduce cross-contamination from previously purified sequences.

LGC's optimized reverse-phase cartridge (RPC) purification method offers a high-value alternative, producing oligos with high purity while minimizing cross-contamination. RPC purification effectively removes truncated species and other impurities, without the contamination risks commonly associated with HPLC.
