Cell Culture Contamination Control

Cell Culture Contamination Control: Elimination

Mycoplasma Elimination

If you are unfortunate enough to find mycoplasma in your cell culture or virus stocks we highly recommend immediate autoclaving and disposal of any contaminated cell cultures. After having discarded the contaminated cells along with the used reagents and cell culture media, we recommend thorough cleaning of the whole laboratory.

However, we appreciate that certain cells and virus stocks are rare or historically significant and might benefit from being cleaned up. With this purpose in mind Minerva Biolabs have developed mycoplasma elimination products (Mynox® and Mynox® Gold) that can eradicate mycoplasma from an already contaminated cell culture with negligible cytotoxicity.

Mynox® is an antibiotic-free formulation for mycoplasma eradication from cell cultures. Its activity is based on a biophysical mechanism and therefore does not allow for the development of resistant strains.

Mynox® Gold is a formulation of the classic Mynox® reagent that has undergone further development and combines a standard antibiotic with the Mynox® ingredients. Achieve 100% eradication of mycoplasma with no cytotoxicity and a low resistance risk.

Mynox®Gold

Mynox®Gold - the most effective reagent against mycoplasma 2 in 1 - Double Strength Mynox®Gold represents the further development of the classical Mynox®, which was known for a very high elimination efficiency and low cytotoxicity Mynox®Gold is a combination of a standard antibiotic and a biological reagent. One application comprises of 4 vials, a starter treatment and three main treatments. The starter treatment kills most of the mycoplasma particles without harming the cells. The main treatment kills all remaining mycoplasma leading to a permanent eradication with efficiency rates up to 100 %. One application can be used to cure one cell culture permanently from mycoplasma burden.

Description

Mynox® Gold

Mynox® Gold represents the further development of the classical Mynox®. Mynox® Gold is a combination of a standard antibiotic and the biological Mynox® reagent. One application comprises of a starter treatment that eliminates most of the mycoplasma particles without harming the cells. The main treatment kills all remaining mycoplasma.

Benefits

No cytotoxicity – cytotoxicity is basically not detectable for most cell types. Most cell lines do not show any changes in morphology during treatment.

Low resistance risk – due to the combination of an antibiotic and a reagent showing a biophysical mode of action directly eliminating the mycoplasma, formation of resistances against Mynox® Gold is most unlikely.

Highly effective – the ratio for a permanent eradication is close to 100 %.

Recommended Use / Scope

Mynox® Gold is intended for research use only. Not applicable for clinical treatment. Not applicable for the direct treatment of enveloped viruses or cells harboring Chlamydia or other bacteria as host systems. The antibiotic portion of the product might affect the integrated microorganisms.

Kit Components

1 application of Mynox® Gold contains 1 vial for the starter treatment and 3 vials for the main treatment. Each vial contains 500 µl as a sterile, ready-to-use solution.

Package Sizes

1 application can be used to cure one cell culture permanently from mycoplasma burden.

  • Cat. No. 10-0201 2 applications
  • Cat. No. 10-0501 5 applications
  • Cat. No. 10-1001 10 applications

Result Evaluation

Elimination success should be verified with a suitable mycoplasma detection system, e.g. Minerva Biolabs’s Venor®GeM Mycoplasma PCR Detection Kits.

Required Consumables

6-well tissue culture plate or small tissue culture petri dish (6 mm Ø)

Regular cell culture plastics

Required Lab Devices

Regular cell culture equipment (incubator, centrifuge, pipettes, etc.)

Shelf Life and Storage

Kit components are maintainable at 2 to 8 °C for at least 6 months.

The cultures were inactivated and stained as described by Flick & Gifford. Untreated cultures had been used as reference (100 %).

Applications & Benefits

Applications

  • For the elimination of mycoplasma partcles without harming cells

Benefits

    • No cytotoxicity - Cytotoxicity is basically not detectable for most cell types. Most cells do not show any changes in morphology during treatment
    • Highly effective - The ratio for a permanent eradication is close to 100%
    • Universal for cells - Mynox®Gold can be used for any cell type without restriction. Only cells harboring Chlamydia or other bacteria as host systems can not be treated. The antibiotic portion of the product might affect the integrated microorganisms
    • Universal for Mycoplasma - Any type of microorganisms belonging to the group of Mollicutes, including the Mycoplasma, Spiroplasma, Acholeplasma, Ureaplasma, etc. can be successfully treated with Mynox®Gold
    • Low resistance risk - Due to the combination of an antibiotic and a reagent showing a biophysical mode of action directly killing the mycoplasma, formation of resistances against Mynox®Gold is most unlikely
    • Format - Mynox®Gold contains 1 vial for the starter treatment and 3 vials for the main treatment. Each vial contains
      500µl as a sterile, ready-to-use solution
    • Interoperable - Mynox®Gold can be used with antibiotic selection agents, for example G418, Blastocyclin, and Hygromycin. It also does not interfere with killer genes turned on by Tetramycin or Doxycyclin
