Description

Locked nucleic acid (LNA), or simply locked analog (LA), is a modified nucleic acid backbone consisting of a bicyclic sugar where the 2'-oxygen is connected to the 4'-carbon via a methylene unit. This additional ring restricts the sugar into a particularly favorable hybridization conformation. As a result, LA oligonucleotides exhibit unprecedented thermal stability towards complementary DNA and RNA.

Applications & Benefits

Usage

Coupling: 3 minute coupling time recommended. Increase oxidation time 3 fold for optimal performance. To avoid any exchange of the iPr-Pac group on the G with acetyl, use the UltraMild Cap Mix A (40-4210-xx/40-4212-xx).

Deprotection: UltraMILD deprotection: 0.05M Potassium Carbonate in Methanol, 4 hours at Room Temperature OR 2 hours at Room Temperature in 30% Ammonium Hydroxide.
