Transcyclooctene (TCO) is a strained alkene that reacts with tetrazine labels in an inverse-electron-demand Diels-Alder cycloaddition (IEDDA) reaction. TCO-tetrazine ligation is ultrafast, selective, and has long-term stability in aqueous solutions. This is a 5'-modifier only and is compatible with a single oxidation cycle with 0.02M iodine.

Usage

  • Coupling: 3 minute coupling time recommended.
  • Deprotection: Ammonium hydroxide for 2 hr at 65°C or as required by nucleobases.
