Lambda DNA Restriction Digestion Analysis	
Restriction enzymes cut DNA at specific 
sequences, producing fragments of varying lengths. Lambda DNA is a 
linear viral genome (~48,502 bp) often used as a model in molecular 
biology. Digestion with different enzymes produces distinct patterns, 
which can be visualized through agarose gel electrophoresis. This kit 
contains the full linear Lambda DNA genome, along with three Lambda DNA 
samples pre-digested with the restriction enzymes HindIII and EcoRI, 
alone or in combination.


Students will analyze DNA fragment patterns 
produced by restriction enzyme digestion of Lambda phage DNA. They will 
compare HindIII, EcoRI, and double digests to compare the cutting sites 
and estimate fragment sizes using a DNA ladder.

DNA samples for 12 gel runs:
- Lambda DNA (120 μl)
- HindIII Lambda Digest (120 μl)

- Double Lambda Digest (120 μl)
- 10K DNA Ladder (120 μl)
 

 
 Storage notes: 
- If kept in the freezer, reagents can be stored for 12 months after receipt. 
 Required equipment:
Gel electrophoresis reagents and plastic tubes are supplied by the user
- Agarose
- Gel electrophoresis buffer (e.g. TBE)
- Fluorescent DNA stain (e.g. SeeGreen™ or GelGreen®)
- Plastic tubes for distributing samples to student groups
 The Learning Lab Companion Kit (KT-1510-01) provides sufficient reagents to prepare and run eight gels when using the blueGel electrophoresis system, as well as plastic tubes to distribute samples to student groups.
