Diluent: Anhydrous Acetonitrile Add fresh diluent to product vial to recommended concentration and swirl vial occasionally over several minutes until product is completely dissolved. (Some oils may require between 5 and 10 minutes.) Use care to maintain anhydrous conditions. In case of transfer to alternate vial type, ensure recipient vial has been pre-dried. Coupling: 15 minute coupling time. DCI activator (30-3150-xx) is recommended. To maintain label yield, carry out the synthesis DMT-ON. |
Deprotection: To maintain label yield as a 5' modifier, treat column with 10% diethylamine in acetonitrile for 2 minutes at room temperature (2x) to remove cyanoethyl protecting groups. Rinse column with ACN and then cleave and deprotect as required by nucleobases. |

TOCOPHEROL LABELLING
Vitamin E is both lipophilic and non-toxic even at high doses so would be an excellent candidate as a lipophilic carrier for oligonucleotides. Therefore, as an addition to our cholesteryl product line, we offer simple a-tocopheryl (vitamin E) labelling. Totally synthetic a-tocopherol is racemic at its three chiral centers and is used to prepare this product.
